The RNA Integrity Number was developed by Agilent Technologies. The algorithm was generated by taking hundreds of samples and having specialists manually assign them all a value of 1 to 10 based on their integrity, with 10 being the highest.

Transcript quantification determines the steady state levels of alternative transcripts within a sample, enabling the detection of differences in the expression of alternative transcripts under different conditions.

Result. We developed the transcript integrity number (TIN) to measure RNA degradation. When applied to 3 independent RNA-seq datasets, we demonstrated TIN is a reliable and sensitive measure of the RNA degradation at both transcript and sample level.

It is generally regarded that a sample with a RNA Integrity Number (RIN) of 10 is indicative of the best quality RNA whereas lower values result from RNA degradation.
