Lynch syndrome I is characterized by an autosomal dominantly inherited proclivity to early onset colonic cancer with proximal predominance and an excess of multiple primary colonic cancer. Lynch syndrome II has all of these features plus extracolonic cancer sites, the most common of which is endometrial carcinoma.

Genetic testing looks for inherited Lynch syndrome mutations. Your doctor may recommend a multi-gene panel, which looks for mutations in several genes at the same time, including the genes associated with Lynch syndrome. The MSH2 gene provides instructions for making a protein that plays an essential role in repairing DNA. This protein helps fix errors that are made when DNA is copied (DNA replication) in preparation for cell division.

Lynch syndrome is due to inherited changes (mutations) in genes that affect DNA mismatch repair, a process that fixes mistakes made when DNA is copied. These genes (MLH1, MSH2, MSH6, PMS2, and EPCAM) normally protect you from getting certain cancers, but some mutations in these genes prevent them from working properly.

Lynch syndrome used to be called hereditary nonpolyposis colorectal cancer (HNPCC). HNPCC is a term used to describe families with a strong history of colon cancer. Lynch syndrome is the term used when doctors find a gene that runs in the family and causes cancer.

What does it mean to test positive for a MLH1, MSH2, MSH6, PMS2, or EPCAM gene mutation? Mutations in the MLH1, MSH2, MSH6, PMS2, and EPCAM genes cause a hereditary cancer predisposition condition called Lynch syndrome, also known as Hereditary Non-Polyposis Colorectal Cancer (HNPCC) syndrome.

If you have a mutation in the MSH2 gene, this means you have a condition called Lynch syndrome. Lynch syndrome increases your risk for certain types of cancers, including: Colorectal (colon and rectal) cancer. Uterine (endometrial) cancer.

Lynch Syndrome Symptoms Bloody stool. Unexplained diarrhea. A long period of constipation. Abdominal cramps. Decrease in size or caliber of stool. Gas pain, bloating, fullness. Unexplained weight loss. Lethargy and vomiting.
