Restriction enzymes are traditionally classified into four types on the basis of subunit composition, cleavage position, sequence specificity and cofactor requirements.

Restriction mapping involves cutting DNA at various different recognition sites. Fragments are then separated and identified with gel electrophoresis. When a plasmid is cut by R.E only one strand of DNA is produced. Because of this. combinations of R.E are used to cut the plasmid into two fragments.
