Adapter ligation—A ligase enzyme covalently links the adapter to the DNA fragments, making a complete library molecule. Adapters serve multiple functions: they can attach the sequences to a flow cell or ensure sequencing platform-specific compatibility.

What is an Adaptor? An adaptor molecule is a double-stranded, chemically synthesised oligonucleotide that is used for the ligation of DNA. The adaptors have one sticky and one blunt end. The blunt end is ligated with the blunt end of the target DNA producing a DNA fragment with sticky ends.

Adapter dimers contain full-length adapter sequences that are able to bind and cluster on the flow cell and generate sequencing data. In contrast, primer dimers do not contain complete adapter sequences, and are not able to bind or cluster on the flow cell, so are not sequenced.

Adapter sequences are short oligonucleotides used to be ligated to the ends of DNA fragments of interest, so that they can be combined with primers for amplification.

Prior to sequencing, DNA or RNA samples are prepared following a library preparation protocol. With any sequencing platform, synthetic adaptors are required to be ligated to the DNA or RNA fragments of interest. These adaptors are recognized by the sequencing platform, allowing sequencing to start.
