Filling out the XF96 Assay Preparation Checklist is essential for ensuring a successful assay run. This guide provides a clear and comprehensive overview of each section of the form, directing you on how to prepare effectively and accurately.
Follow the steps to complete the XF96 assay preparation checklist online.
- Click the ‘Get Form’ button to acquire the XF96 Assay Preparation Checklist and open it in your preferred editor.
- Enter the date at the top of the checklist to document when the assay preparation will occur.
- Fill in the lab technician's name to identify who is responsible for the assay preparation.
- Specify the cell type you will be using for the assay, as this is crucial for accurate results.
- Review the status and tasks outlined under the 'Night before' section. Ensure you prepare the unbuffered assay medium with a pH of 7.4 at 37°C, hydrate the cartridge for a minimum of 12 hours in a non-CO2 incubator, and turn on the XF Analyzer and controller to allow them to reach 37°C.
- Create or select a protocol for the assay from an existing template.
- On the day of the assay, warm the assay medium to 37°C in a non-CO2 environment before proceeding.
- Prepare injectable compounds using the previously warmed assay medium, adjusting the pH to 7.4 at 37°C.
- Examine the cells under a microscope to ensure they are ready for the assay.
- Replace the cell culture medium with the prepared assay medium to start the assay process.
- Place the cells in the assay medium into a non-CO2 incubator for at least 30 minutes before the assay run.
- Load the injectable compounds into the designated ports (A, B, C, D) of the cartridge.
- Insert the cartridge and utility plate with calibration buffer into the XF Analyzer.
- Run the calibration protocol from the template to ensure accuracy of the cartridge measurements.
- After calibration, check the cells under the microscope again.
- Upon successful calibration, load the cell culture plate into the XF Analyzer.
- Continue executing the assay protocol as per the instructions provided in your protocol template.
- Utilize the notes section at the bottom of the checklist to record any observations or important information.
