Fungus culture identification uses various methods, including microscopic morphology, biochemical reactions, growth temperature and culture characteristics. MALDI-TOF and Molecular methods may be used when needed.

Fungi are usually classified in four divisions: the Chytridiomycota (chytrids), Zygomycota (bread molds), Ascomycota (yeasts and sac fungi), and the Basidiomycota (club fungi). Placement into a division is based on the way in which the fungus reproduces sexually.

The identification of fungi using molecular techniques is carried out by the sequencing of PCR amplified part of 18S rRNA genes with universal primers to fungal species (Monod et al., 2005; Hensel and Holden, 1996).

Characteristics of Fungi Fungi are eukaryotic, non-vascular, non-motile and heterotrophic organisms. They may be unicellular or filamentous. They reproduce by means of spores. Fungi exhibit the phenomenon of alternation of generation. Fungi lack chlorophyll and hence cannot perform photosynthesis.

Sequencing Independent Methods. DNA sequencing of the internal transcribed spacer (ITS) and large subunit (LSU) regions of rRNA, followed by comparative sequence analysis, has been the 'gold standard' for molecular identification of most fungi, particularly of culturable fungi.

The kingdom Fungi contains five major phyla: Chytridiomycota, Zygomycota, Ascomycota, Basidiomycota, and Glomeromycota.
