A validated method for neutralizing the antimicrobial properties of a product must meet two criteria: neutralizer efficacy and neutralizer toxicity.

AETs are performed by challenging the product with a panel of microorganisms to verify the preservative's efficacy against possible contamination agents. A typical study may include gram-positive cocci, gram-negative fermentative bacilli, gram-negative non-fermentative bacilli, yeast, and mold.

A variety of laboratory methods can be used to evaluate or screen the in vitro antimicrobial activity of an extract or a pure compound. The most known and basic methods are the disk-diffusion and broth or agar dilution methods. Other methods are used especially for antifungal testing, such as poisoned food technique.

The AET (also called Preservative Efficacy Test) is performed to determine if the antimicrobial(s) or preservative(s) in nonsterile or sterile multi-dose aqueous products can effectively prevent the proliferation of microorganisms.
