There are two types of NMR spectrometers, continuous-wave (cw) and pulsed or Fourier-Transform (FT-NMR).

How Does NMR Actually Work? When molecules are placed in a strong magnetic field, the nuclei of some atoms will begin to behave like small magnets. If a broad spectrum of radio frequency waves are applied to the sample, the nuclei will being to resonate at their own specific frequencies.

Several different NMR-sensitive nuclei can be used in the study of biological systems, and the most common are 1H, 13C, 31P, and 19F. Compared to 1H, the other NMR nuclei provide substantially less signal and consequently, sensitivity is for most of them even a greater challenge than for 1H MRS.

NMR spectroscopy is the use of NMR phenomena to study the physical, chemical, and biological properties of matter. Chemists use it to determine molecular identity and structure. Medical practitioners employ magnetic resonance imaging (MRI), a multidimensional NMR imaging technique, for diagnostic purposes.

H NMR is the go-to technique to help identify or confirm the structure of organic compounds or those that contain protons. A solution-state proton spectrum is relatively fast to acquire, compared with other nuclei, and a lot of information about the structure of a compound can be deduced from it.

Some of the peaks are isolated (a singlet), some are two peaks close together (a doublet), others have three peaks (a triplet), etc. Each set of peaks contains three types of information: chemical shift, integration, and multiplicity (also called splitting).

The types of NMR usually done with nucleic acids are 1H or proton NMR, 13C NMR, 15N NMR, and 31P NMR.

Each signal in a proton NMR spectrum may or may not be split into one or more peaks. This is called signal multiplicity and gives rise to names such as singlet, doublet, triplet, quartet, pentet, and multiplet. The most common concept associated with signal multiplicity is the n+1 rule.
