Membranes are composed of lipids, proteins and sugars This structure is generally referred to as the phospholipid bilayer. In addition to the various types of lipids that occur in biological membranes, membrane proteins and sugars are also key components of the structure.

In 1890, an update to the Cell Theory stated that cell membranes existed, but were merely secondary structures. It was not until later studies with osmosis and permeability that cell membranes gained more recognition. In 1895, Ernest Overton proposed that cell membranes were made of lipids.

The fundamental structure of the membrane is the phospholipid bilayer, which forms a stable barrier between two aqueous compartments. In the case of the plasma membrane, these compartments are the inside and the outside of the cell.

The first forms of cellular life required self-assembled membranes that were likely to have been produced from amphiphilic compounds on the prebiotic Earth. Laboratory simulations show that such vesicles readily encapsulate functional macromolecules, including nucleic acids and polymerases.

All of the lipid molecules in cell membranes are amphipathic (or amphiphilic)—that is, they have a hydrophilic (“water-loving”) or polar end and a hydrophobic (“water-fearing”) or nonpolar end. The most abundant membrane lipids are the phospholipids. These have a polar head group and two hydrophobic hydrocarbon tails.

ing to Danielli and Davidson, 1935, plasmalemma is a double phospholipid layer surrounded on either side by a layer of hydrated globular proteins in the form of "Protein-Lipid-Lipid-Protein". The hydrophobic or non-polar tails of the two lipid layers are towards the centre.

Membranes Form Spontaneously The head end is attracted to water, while the tail end moves away from water. When phospholipids are placed into water, they organize themselves into a structure called a bilayer. The water-fearing tail regions clump together on the inside of the bilayer, as far from water as possible.
