Thin layer chromatography is an important tool for detecting amino acids by variety of spray reagents. Among these ninhydrin is the most popular due to its high sensitivity. However, ninhydrin produces the same purple/violet color with most amino acids.

Bromothymol blue is incorporated into the mobile phase as indicator and thus the acids appear as yellow spots on a blue background.

Preparation: Solution A: 1.7 g basic bismuth nitrate in 100 ml water/acetic acid (4:1). Solution B: 40 g potassium iodide in 100 ml of water. Mix reagents together as follows: 5 ml A + 5 ml B + 20 ml acetic acid + 70 ml water. Spray plates, orange spots develop.

However, if the substance is colourless, it may be treated with a reagent, which imparts it a characteristic colour. This reagent is given the name developer. Iodine is the most commonly used developer in paper chromatography. Several other techniques are available for locating the spots.

Detection is performed by measuring absorbance of the column eluate with an ultraviolet detector at 280 nm, the absorbance wavelength for tyrosine and tryptophan, or at 214 nm, the absorbance wavelength for most amino acids and the peptide bond.

After the chromatogram has dried, one can identify the spots. If the spots can be seen, outline them with a pencil. Sometimes the color will fade over time, so the pencil outline is important. If the spots are not obvious, the most common visualization technique is to hold the paper under an ultraviolet lamp.

The most common non-destructive visualization method for TLC plates is ultraviolet (UV) light. A UV lamp can be used to shine either short-waved (254nm) or long-waved (365nm) ultraviolet light on a TLC plate with the touch of a button.
