The origin of miRNA-biology lies in the evolution of a mechanism that allowed for processing of long endogenous transcripts into short RNA duplexes that are further recognized and cleaved by Dicer.

miRNAs were discovered in 1993 by Lee and colleagues93 in the nematode Caenorhabditis elegans. In these organisms, the downregulation of LIN-14 protein was found to be essential for the progression from the first larval stage (L1) to L2.

miRNAs are small non-coding RNAs, with an average 22 nucleotides in length. Most miRNAs are transcribed from DNA sequences into primary miRNAs (pri-miRNAs) and processed into precursor miRNAs (pre-miRNAs) and mature miRNAs.

Novel microRNAs can originate from the random formation of hairpins in "non-coding" sections of DNA (i.e. introns or intergene regions), but also by the duplication and modification of existing microRNAs.

These miRNAs can be located within the introns of protein coding genes or within long noncoding RNAs. They can also be classified into novel families based on the identity of their host genes (Rodriguez et al., 2004).

microRNA is the name of a family of molecules that helps cells control the kinds and amounts of proteins they make. That is, cells use microRNA to help control gene expression. Molecules of microRNA are found in cells and in the bloodstream.

The expression of miRNAs can be regulated by epigenetic modification, DNA copy number change, and genetic mutations. miRNAs can serve as a valuable therapeutic target for a large number of diseases.

Origin: The siRNA is an exogenous double-stranded RNA uptaken by cells, while miRNA is single-stranded and comes from endogenous non-coding RNA. Besides, the siRNA is present in lower animals and plants, but not found in mammals; whereas miRNAs are present in all the animal and plant.
