The ability of NK cells to be very rapidly activated by inflammatory cytokines, to secrete effector cytokines, and to kill infected or stressed host cells, suggests that they may be among the very early responders during infection.

Natural killer or NK cells are a sub-population of Large Granular Lymphocytes (LGLs) which arise from a common NK/T-cell progenitor. Accumulating evidence indicates that NK cells can develop and mature both in the bone marrow and secondary lymphoid tissues (SLTs) including tonsils, spleen, and Lymph nodes (LNs)[1].

NK cells are derived from the CLP, which differentiates into a heterogeneous pre-NKP/ILCP population distinguished from the NKP by its expression of IL-7R and lack of CD122 expression. From the NKP, cells begin to express NK cell markers NK1.

The human killer cell immunoglobulin-like receptors (KIR; also known as CD158) are a family of transmembrane glycoproteins expressed on NK cells and a subset of T cells. The KIR are key regulators of the development, tolerance and activation of NK cells.

In 1975, Rolf Keissling and colleagues in Sweden described a similar natural cytotoxicity and determined that the killer cell was genetically regulated, and killed many types of tumours. Keissling named the effector cell a "natural" killer (NK) cell.
