It is known that the accumulation of glucose and cellobiose, end products of hydrolysis, inhibit cellulases and decrease glucose yields. Aside from these end products, however, other monosaccharides such as mannose and galactose (stereoisomers of glucose) decrease glucose yields as well.

Oligomeric phenolics could inactivate cellulases by reversibly complexing them. Simple and oligomeric phenolics could also inhibit enzymatic cellulolysis by adsorbing onto cellulose. Individual cellulases showed different susceptibility toward these inhibitions.

Glucose and cellobiose manifested competitive inhibition with almost comparable inhibition constants (Ki) of 24.7 and 26.3 mM respectively, while Vmax = 0.26 g L−1h−1. On the contrary, with galactose and mannose, the inhibition constants decrease to 10.9 and 11.1 mM respectively.

Cellulase consists of three enzymes: β-glucosidase, endo-1,4-β-D-glucanase (endoglucanase) and exo-1,4-β-D-glucanase (exoglucanase). These three enzymes are involved in the hydrolysis of cellulose by synergetic action for accomplished and effective hydrolysis of cellulose [7].

Cellulase enzymes are used to break down the cellulose of plant cell walls into simple sugars that can be transformed (fermented) by microbes to fuels, primarily ethanol, as well as to chemicals, plastics, fibers, detergents, pharmaceuticals, and many other products.

There are two catalytic mechanisms of cellulases. They are simply introduced as retaining mechanisms and inverting mechanisms. Cellulases cleave glucosidic bonds by using acid-based catalysis. The hydrolysis is performed by two catalytic residues of the enzyme: a general acid (proton donor) and a nucleophile/base.

Cellulase enzymes are used to break down the cellulose of plant cell walls into simple sugars that can be transformed (fermented) by microbes to fuels, primarily ethanol, as well as to chemicals, plastics, fibers, detergents, pharmaceuticals, and many other products.

Transcription of the cellulase genes is strictly inhibited by glucose, and is induced by cellulose or disaccharides (for example, sophorose, lactose, and gentiobiose) [6], [7].
